چکیده

Computer-aided drug design (CADD) shift toward using quantum-mechanics (QM)-based approaches is not only the result of the ever growing computational power but also due to the need for more accurate and more informative approaches to describe molecular properties and binding characteristics than the currently available ones. QM-approaches do not suffer from the limitations inherent to the ball-and-spring description and the fixed atom-centered charge approximation in the classical force fields mostly used by CADD methods.[1] We introduce a protocol for shifting 3D-QSAR, one of the most widely used ligand-based drug design approaches, through using QM-based molecular interaction fields (MIFs) which are the electron density (r), hydrogen bond donor field (HDF), hydrogen bond acceptor field (HAF) and molecular lipophilicity potential (MLP) to overcome the limitations of the current force-field-based MIFs (FF-MIFs). The average performance of the

Application of 3D-QSAR on a Series of Potent P38-MAP Kinase Inhibitors

One of the most applied methods in drug industry for development of new drugs is 3D-QSAR methodology. As p38-mitogen-activated protein kinase (p38-MAPK) plays a crucial role in regulating the production of such proinflammatory cytokines as tumor necrosis factor-α (TNF-α) and interleukin-1, emerging as an attractive target for new anti-inflammatory agents, we used a 3D-QSAR based method of Compa...

3D-QSAR Modeling of Anti-oxidant Activity of some Flavonoids

The anti-oxidant activities for a diverse set of flavonoids as TEAC (Trolox equivalent anti-oxidant capacity), assay were subjected to 3D-QSAR (3 dimensional quantitative structural-activity relationship) studies using CoMFA (comparative molecular field analysis) and CoMSIA (comparative molecular similarity indices analysis). The obtained results indicated superiority of CoMSIA model over CoMFA...
